Purification of Chitinase from Pseudomonas vesicularis KW-15

Abstract

|
The optimum cultural temperature and time for the production of chitinase from Pseudomonas vesicularis KW-15 were 30t and 60 hrs, respectively. The enzyme was purified by ammonium sulfate fractionation, affinity adsorption, two times of DEAE-cellulose column chromatography. Specific activity of the purified enzyme was 11.0 U/mg. protein and the yield of the enzyme activity was 16.2%. The purified enzyme showed a single protein band on polyacrylamide gel electrophoresis and its molecular weight was estimated to be 62,000 by SDS-polyacrylamide disc gel electrophoresis. The isoelectric point of the purified enzyme was pH 4,9.
